Osteoporotic fractures and osteoarthritis (OA) are on an upward trajectory with an increasing aging population in modern societies, costing the United States healthcare system an estimated annual of USD57 billion1 and USD65.5 billion2 respectively.

Bone Disorder

Decreased bone mass and qualitative change in the bone micro-architecture associated with increased fracture risk are characteristics of osteoporosis. The major complication of osteoporosis is osteoporotic fractures that result in limited ambulation, chronic pain, loss of independency and low quality of life, which eventually increases the risk of mortality.

Causes of osteoporosis

  • Decreased MSCs in bone tissue due to homing disorder
  • Less mature osteoblasts as a result of impaired osteogenic ability of MSCs
  • Imbalance of osteoblasts and osteoclasts aggravated by the senescence of MSCs.
  • Immune disorders in the bone niche caused by abnormal activation of immune cells and impaired immunoregulatory ability of MSCs.

Damaged Joint

Eighty-five percent of osteoarthritis cases are in the knee, followed by the hand and hip. One major cause of OA is obesity with excess weight loading pressure on weight-bearing joints. Abnormal loading of cartilage eventually causes damage to the cartilage matrix and triggers inflammation.

MSC-based therapy initiates repair process

MSC transplantation into the bone tissue and joint has treatment potential for osteoporosis4 and OA5 albeit limited efficacy.
